fa نگهداری بذر Satureja spicigera در شرایط فراسرد Cryopreservation of Satureja spicigera seeds تخصصي Special پژوهشي Research در سالهای اخیر، در کشور ما نیز توجه زیادی به گیاهان دارویی شده است. از آنجا که گونههای مختلف گیاه مرزه بر اثر برداشت بی‌رویه از عرصه‌های طبیعی توسط انسان و تنش‌های زنده و غیرزنده تهدید شده و کشت کار پیدرپی یک گونه باعث کاهش تنوع ژنتیکی آنها می‌گردد، حفظ ذخایر ژنتیک این گونه‌ها از اهمیت بالایی برخوردار است. با استفاده از تکنیک نگهداری بذر در دمای فراسرد که یکی از روشهای نگهداری ژرمپلاسم در شرایط خارج از رویشگاه است، میتوان بذر را به طور طولانی مدت، با هزینه بسیار کمتر و بدون از دست دادن قوهنامیه ذخیرهسازی کرد. برای نگهداری بذر Satureja spicigera در شرایط فراسرد، از پیش تیمارهای گلیسرول، محلول ویتریفیکاسیون گیاهی Plant Vitrification Solution 2 PVS2)) و کاهش رطوبت بذر قبل از ورود به ازت مایع استفاده شد. بذرهای تیمار شده به مدت یک هفته، یک ماه و سه ماه در دمای -196 ºC نگهداری شدند. در این تحقیق شاخصهای جوانهزنی و رشد (درصد جوانهزنی، سرعت جوانه زنی) بذر ارتودکس گونه مرزه S. spicigera در شرایط فراسرد به مدت یک هفته، یک ماه و سه ماه مورد ارزیابی قرار گرفت. نتایج نشان داد که بین پیشتیمارهای مختلف، از لحاظ شاخص جوانهزنی و سرعت جوانهزنی اختلاف معنیداری وجود دارد و میتوان با استفاده از فناوری فراسرد و بکارگیری پیش‌تیمار مناسب، بذر این گونه ارزشمند و در حال خطر را، برای مدت زمان بسیار طولانی حفظ و از انقراض این گونه منحصر به فرد جلوگیری نمود. Using seed cryopreservation, as one of the Ex situ plant germplasm conservation method we can store seed for long-term, with much lower expenses and without losing seed viability. In order to preserve Satureja spicigera seed under -196ºC condition, three pre-cryopreservation treatments, PVS2 solution, Desiccation and Glycerol were applied. The treated seeds were transferred into Liquid Nitrogen (LN) and stored for 7, 30 and 90 days respectively. In this study the effect of cryopreservation on germination and growth indices (germination percent, germination rate, seed vigour index, plantlet length on orthodox seeds of plant species (Satureja spicigera) in storage conditions of cryopreservation (-196ºC) were evaluated for 7, 30 and 90 days. Comparing treatments, significant differences were observed on germination indices and germination rate.
