Wheat Proteomics under Drought Stress: From Signaling Networks to Metabolic and Proteostatic Reprogramming for Sustainable Production

Drought stress can be considered one of the key environmental factors impeding the efficiency of world wheat production by affecting its productivity through the disruption of photosynthetic process, energy balance, and redox homeostasis perturbations. Given that proteins can be seen as ultimate gene expression products and the key regulators of biological processes, proteomic approaches in combination with transcriptomic and metabolomic studies provide a detailed picture of adaptation mechanisms in response to the adverse effect of drought in wheat. The present review considers modern knowledge about the role of proteomics in studying drought response in wheat from a comprehensive standpoint and provides the classification of results obtained in accordance with regulatory axes, such as advanced signaling networks, redox homeostasis regulation, antioxidant systems, proteostasis, and alteration of metabolic reprogramming. There is ample evidence about the involvement of ABA-, calcium-, and MAPK-dependent pathways, which result in the activation of defensive chaperones (HSPs) and enhanced ubiquitin-proteasome system to facilitate the destruction of damaged proteins. Importantly, changes in the expression of ROS scavengers SOD, CAT, and APX contribute to energy balance and oxidation resistance. Overall, the combination of multiple omics approaches along with high throughput precision phenotyping, particularly in field-based experiments, acts as an excellent approach that can be employed to discover reliable biomarkers. Such an interdisciplinary approach becomes crucial in order to promote the development of durable drought tolerant cultivars, thus contributing to sustainable wheat production.
